Myeloprotection with activated carbon in doxorubicin-treated rats

Chemotherapy can often cause a variety of side effects including bone marrow (BM) suppression, termed as myelosuppression. Accordingly, facile and effective management chemotherapy-induced myelosuppression is currently pivotal task for experimental pathologists oncologists. Here, we chose to use activated carbon (AC) with an extensive surface area studying its possible protective effectiveness respect BM in doxorubicin (DOX)-treated rats. Spherical AC extended up 4490 m2/g was prepared per os (p/o) delivery, whereas intraperitoneal (i/p) delivery used the powdered form that derived from aforementioned spherical AC. During monthly treatment animals DOX these two components were delivered alternately (not same day). After treatment, cells isolated femurs sacrificed animals, stained acridine orange (AO) analyzed by flow cytometry. Regardless route (p/o or i/p), apparent myeloprotection regenerative effect observed received DOX, evidenced recovery populations total nucleated (TNC) polychromatic (immature) erythrocytes accompanied considerable reduction number apoptotic/dead among TNC (≤2.0%). Moreover, result administrations, there significant increase AO green far-red fluorescence intensities population TNC, which suggestive ongoing conformational changes chromatin DNA associated cell proliferation. Thus, preparations under present conditions effectively tackle DOX-induced via mechanisms not necessarily adsorptive detoxification.
